Spatial analysis for visualizing small RNA species implicated in disease and development

Outline:
The RNAscope™ in situ hybridization (ISH) technology provides a powerful method to detect gene expression within the spatial and morphological tissue context. The proprietary probe design in combination with advanced signal amplification enables robustly high signal-to-noise and allows for the detection of gene transcripts at the single molecule level with single-cell resolution.
Now, this webcast will present the miRNAscope™ Assay, an advanced in situ hybridization platform that allows for highly specific and sensitive visualization of small RNA species expression with single-cell resolution.

ASO
ASOs are used as therapeutic solutions for mod- ulating disease processes by altering synthesis of a specific protein.
Example detection of ASO target (yellow arrows) in the treat- ed mouse quad muscle tissue samples and not in the vehicle
samples indicates successful delivery of ASO therapy.

siRNA
siRNAs are being developed as potential thera- peutic agents by silencing specific gene targets.
Example of specific detection of the siRNA#1 probe (yellow arrows) in siRNA#1 treated mouse liver tissue samples only and not in the negative saline nor siRNA#2 treated samples indicates success- ful delivery of siRNA therapy and specificity of the miRNAscope Assay. Biodistribution of siRNA target is also observed in the siRNA#1 treated samples.
