It is well known that the major histocompatibility complex (MHC) region of the human genome is 4.5 Mb long and is one of the most complex and difficult to study regions of the human genome. Our speaker Prof. Yun Liu and his team used a combination of CRISPR-based targeted enrichment strategy + 10X genomics and PacBio HiFi sequencing to detect enrichment with only 20ng of DNA input. The captured regions were subjected to 30-fold HiFi sequencing.
