谷氨酰胺合成酶（GS）可以促進谷氨酰胺的合成進而促進蛋白質和核苷酸的合成以及腫瘤細胞生長，因此GS通常被認為是促癌的。然而在有β-Catenin激活的小鼠肝癌模型中敲除肝臟GS卻顯著促進了肝腫瘤生長。在機理方面，在肝臟中特異性敲除GS加劇了高氨血症並促進了非必需氨基酸合成，進而激活了mTORC1。 mTORC1和谷氨酸轉氨酶的藥物抑制和肝臟特異性敲除均抑制了GS敲除促進的肝腫瘤生長。因此，GS介導的氨清除在存在β-Catenin激活突變的肝臟中可抑制肝腫瘤生長。...
